Role and mechanism of endothelin-1 in the deterioration of visual function of glaucomatous eyes

AIM: To probe into the role and mechanism of ET-1 in the deterioration of visual function in the patients with glaucoma in order to provide a new approach to the prevention and cure of glaucoma from molecular biology. METHODS: The experiment was completed in the central laboratory of the Affiliated Hospital of Guangdong Medical College from July 2002 to June 2003. According to the deterioration degree of visual function and the area ratio of rim to disc, the ET-1 level in the plasma was determined by radioimmunoassay in 16 patients with early glaucoma, 21 with glaucoma progressive glaucoma, 18 with terminal glaucoma and 30 normal adults. RESULTS: The average ET-1 level in the plasma were (82.17 ± 14.88), (65.35 ± 18.45), (69.07 ± 25.28), (91.23 ± 13.26) and (107.06 ± 34.02) mg/L respectively in glaucoma group, control group, the deterioration of visual function in early, progressive and terminal periods, and the level of ET-1 in deterioration of visual function in early period was higher than that in control group, but without significant difference (t = 3.98, P > 0.01). The level of ET-1 in the deterioration of visual function in progressive or terminal period were either significantly increased as compared with that in the control group(t = 4.62, t = 4.89, P < 0.01). The ET-1 level was significantly increased in the deterioration of visual function in progressive and terminal period as compared with that in early period (t = 4.37, t = 4.53, P < 0.01). In the deterioration of visual function in early, progressive and terminal periods, the average area ratio of rim to disc was 0.43 ± 0.11, 0.76 ± 0.23, 0.94 ± 0.21. The area ratio of rim to disc and the level of ET-1 was positively correlated(r= 0.912 4, P < 0.001). CONCLUSION: The increased ET-1 level in the plasma of glaucoma may be highly related to the deterioration degree of visual function in patients with glaucoma. LA: Chinese
