The effect of MSeA on the secretion of MMP-2 and TIMP-1 in cultured bovine trabecular cells

AIM: To understand the effect of methyl seleninic acid (MSeA) on the secretion of matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in cultured bovine trabecular cells and to investigate the role of MSeA in the pathogenesis of primary open-angle glaucoma (POAG). METHODS: TM cells were incubated in DMEM containin g 1, 2, 5, 10μg/L MSeA or DMEM without any experimental reagent for 24h, respectively. The expression of MMP-2 were assayed by immunohistochemistry method and TIMP-1 were assayed by Western blot. RESULTS: MSeA could decrease the expression of MMP-2 and TIMP-1. CONCLUSION: MSeA can affect the balance of extracellular matrix (ECM) by changing the secretion of MMP-2 and TIMP-1, which contributes to the pathogenesis of POAG. LA: Chinese
