Establishment of chronic glaucoma model by translimbal laser photocoagulating trabecular meshwork

OBJECTIVE: To establish a simple, economic and practical chronic experimental glaucoma model has an important significance in the basic study and clinical research of glaucoma. The purpose of this paper was to creat a chronic glaucoma model by laser irradiating trabecular meshwork (TM) via external limbus and assess the damage of retinal ganglion cells. METHODS: Krypton laser was used to target at the TM in the right eyes of 20 Wistar rats with the wavelength 532 nm, light spot diameter 200 μM, duration 0.7 s and power 0.4 W. The left eyes without treatment were as control group. 80-100 spots were set along the 360(degrees) of eyeball for 2 times at a 7-day interval. Intraocular pressure (IOP) was measured using a tonometer at different times following photocoagulation of laser. The average density of retinal ganglion cells (RGCs) in photocoagulated eyes and control eyes was compared. RESULTS: IOP in the photocoagulated eyes were (25. 30 ± 9.40), (27.15 ± 6.02), (26.12 ± 8.63), (26.20 ± 2.69), (26.15 ± 3.29) and (25.55 ± 1.20) mmHg at 3 days after the first laser and at 3, 7, 14, 28, 35 days after the second laser respectively. IOP in the control eyes were (12.25 ± 3.54), (12.30 ± 3.48), (12.28 ± 3.76), (12.14 ± 3.53), (12.36 ± 3.72) and (12.60 ± 3.49) mmHg, showing that IOP of experimental eyes at various times after laser were significantly higher than that before laser (P < 0.01). The average density of RGCs in the photocoagulated eyes was (1 992 ± 108) cells/mm2, and that of the control eyes was (2516 ± 196) cells/mm2, indicating a significantly lower density value in the photocoagulated eyes in comparison with control eyes at 35 days after the second irradiation (P < 0.01). CONCLUSION: Photocoagulation of TM with translimbal laser to elevate the IOP is a simple and an effective method of establishing chronic experimental glaucoma model. LA: Chinese
