A report of a SNP EX3 1209A>G of prostaglandin F receptor gene among Malaysians

OBJECTIVE: To report the detection of the SNP located in the untranslated region of exon 3 in Prostaglandin F receptor gene, EX3 1209A > G, among Malaysian population. DESIGN: A prospective cohort study. Materials and METHODS: 54 subjects consisted of 34 glaucoma patients and 20 normal controls were enrolled into this study. DNA was extracted from blood using commercial kit. DNA was subjected to PCR amplification and then, samples were screened using denaturing High Performance Liquid Chromatography (dHPLC). DNA sequencing was performed to confirm the mutation. RESULTS: Out of 54 subjects screened with dHPLC, 11 subjects revealed a heterozygous peak. Sequencing analysis confirmed that there was a substitution of A to G at position EX3 1209A>G. The allele frequencies of SNP EX3 1209A > G was A = 0.898 and G = 0.102. CONCLUSION: dHPLC proved to be a powerful tool in mutation screening despite small sample size. The allele frequencies for EX3 1209A > G seems to be in agreement with other reported populations. A study to associate this SNP in a group of glaucomatous patients to the response to an antiglaucomatous drug (Latanoprost) is underway.
