As diseases of the 2nd and 3rd retinal neurons, congenital stationary night blindness (CSNB) 1 and 2, melanoma-associated retinopathy (MAR), and glaucomatous optic neuropathy have been studied extensively and their clinicopathological aspects have been clarified. In fact, causative genes responsible for CSNB 1 and 2 have been identified as NYX and CACNA1F genes, respectively. In terms of MAR, immunohistochemical analysis has revealed that patients' serum specifically reacts with retinal bipolar cells. Thus, this is the solid proof that MAR is the 2nd retinal neuron disease. As the major disease affecting the 3rd retinal neuron, glaucomatous optic neuropathy is known to be caused by both intraocular pressure (IOP) dependent and independent factors. Our group has been engaged in research into the IOP-independent causative factors and we have found that autoimmune mechanisms and glutamate toxicity in the glaucomatous optic neuropathy etiology are contributory factors.
