AIM: To observe the change of superoxide dismutase (SOD) and malondiadehyde (MDA) in the retina of rabbits with chronic ocular hypertension (OHT) and the protective effect of crocus sativus. METHODS: Twenty-four rabbits were randomly divided into four groups: control, OHT, and two crocus sativus-treated groups (n = 6, totally 12 eyes for each group). Chronic ocular hypertension in OHT group and two crocus sativus-treated groups was induced by injecting 0.2mL of 3g/L Carbomer solution into anterior chamber. The rabbits in two treated groups were treated with extract preparation of crocus sativus every day, 20mg/kg and 80mg/kg respectively. The animals were sacrificed at 7 or 14 days after Carbomer injection and the contents of superoxide dismutase (SOD) and malondiadehyde (MDA) in retina were assayed and compared among groups. RESULTS: The level of SOD in retinal tissue in OHT and treated groups were markedly lower than that in the control group on day 7 after modeling (P<0.05). The SOD values of two treated groups were obviously higher than that in OHT group and SOD level in the treated group with 80mg/kg crocus sativus was higher than that in the group treated with 20mg/kg crocus sativus (P < 0.05). The level of MDA in retinal tissue in OHT and treated groups were markedly higher than that in the control group at day 7 after modeling (P < 0.01). The MDA values of two treated groups were lower than that in OHT group (P < 0.01) but still higher than the control (P < 0.01). Fourteen days after modelling, the level of SOD in OHT group and two treated groups was comparable or near to that in control group, whereas MDA level was further increased and higher than that in control group (P < 0.01). CONCLUSION: Crocus sativus can effectively enhance the activity of SOD and reduce the content of MDA in retina in early period of OHT. Crocus sativus is an excellent antioxidant which can protect retina against early damage of free radicals. LA: Chinese
