Protection of Nimodipine on optic nerve in the patient with chronic intraocular hypertension

OBJECTIVE: To investigate the optic-neuroprotective effects of Nimodepine (NMD) on chronic intraocular hypertensive eye. METHODS: The chronic intraocular hypertension model was created in 32 rabbits by the injection of 0.2 mL Compound Carbomer solution into anterior chamber. After 3 days of the injection, 0.3% NMD ophthalmic ointment was topically used on the right eyes of rabbit models twice per day for 4 weeks in group A1, and the matrix of 0.3% NMD ophthalmic ointment was used in the left eyes of models at the same way in group A2. Other 16 normal rabbits were used as normal control group. The histological changes of optic nerve and retina were observed under the light microscope. RESULTS: Only a few cells lossing in retinal inner nuclear layer and outer nuclear layer, and lightly vacuolus degeneration was observed under the light and electron microscope, and ultrastructure of retinal cell was normal under the electron microscope in group A1. There was a obvious difference between group A1 and A2. The thickness of retinal inner nuclear layer in group A1 was (25.31 ± 11.01) μm, and that of group A2 was (19.64 ± 10.22) μm with a statistically significant difference between them (P < 0.05). The mean diameter of optic nerve fiber was (1.215 ± 0.064) μm in group A1, and that of group A2 was (1.504 ± 0.094) μm, showing a evident difference between two groups (P < 0.01). The nerve fiber count of optic nerve in group A1 (68 655 ± 9 572) was more than that of group A2 (48 524 ± 17 312) (P < 0.01). CONCLUSION: The topically administration of 0.3% NMD ophthalmic ointment can relieve the damage of optic nerve and retinal inner nuclear layer induced by chronic intraocular hypertension. NMD eye-preparation plays a optic-neuroprotective role on optic nerve and retinal ganglion cells under the chronic intraocular hypertension condition. LA: Chinese
