AIM: To study the effect of cyclosporine A (CsA DDS) on the scarring prevention of the filtering passage during glaucoma filtering surgery. METHODS: Trabeculectomy was performed in 12 New Zealand white rabbits. The left eyes (experimental group) were treated with sustained-release implant of CsA DDS, and the right eye (control group) were treated with the implantation of blank vector during surgery. Intraocular pressure (IOP), filtering bleb and anterior chamber flare were observed after surgery. Aqueous humor samples were gathered and the concentration of CsA in the aqueous humor was measured. Rabbits were killed on the 7th, 14th and 28th day postoperatively in batch and histopatholoical examination was carried out. RESULTS: CsA DDS could be well tolerated in rabbit eyes. The average levels of filtering blebs in experimental eyes had no significant difference compared with control eyes at 3 days after surgery, but were significantly higher than those in controls at 7, 15 and 28 days after surgery. The IOP were significantly different between two groups at 3, 7, 15, 21 and 28 days after surgery. The concentration of CsA decreased slowly with time. HE staining showed 25% obstruction of biting site in experimental eyes and 100% obstruction in controls. CONCLUSION: CsA DDS is a safe and effective treatment to prevent scarring after glaucoma filtering surgery. LA: Chinese
