AIM: To investigate the distribution and expression of RhoA in the rat retina after acute high intraocular pressure. METHODS: Totally 56 SD rats were equally divided into 4 groups; normal group, 1 day, 3 days, 7 days group after establishing acute high intraocular pressure models. The distribution of RhoA in the retina was assayed by immunohistochemistry and the expression was analyzed by semi-quantitative RT-PCR. RESULTS: In normal retina and 1 day after acute high intraocular pressure (AHIOP), RhoA was mainly distributed in retinal ganglion cell layer (GCL). In the group of 3 days after AHIOP, it existed in GCL and inner plexiform layer (IPL). However, it was detected not only in GCL and IPL but also in the inner nuclear layer (INL) and outer plexiform layer (OPL) in the group of 7 days after AHIOP. RT-PCR showed that expression of RhoA mRNA was minimal. However, the expression of RhoA in three groups of AHIOP was increased significantly than normal group (P < 0.05). CONCLUSION: The distribution and expression of RhoA are extended and enhanced after AHIOP, RhoA play an important rule in the course of inhibiting axon regrowth. LA: Chinese
