A study of experimental carbomer glaucoma and other experimental glaucoma in rabbits

OBJECTIVE: To establish models of experimental glaucoma in rabbits to investigate the treatment of glaucoma. METHODS: Twenty-one normal albino rabbits weighing 2.5-3.0 kg were studied. The rabbits were randomly divided into four groups: (1) anterior chamber injection of 0.1 ml 0.3% carbomer; (2) 0.2 ml methylcellulose; (3) 0.2 ml compound methylcellulose; and (4) subconjunctival injection of 5 mg dexamethasone, were performed on them respectively to induce glaucoma. The intraocular pressure (IOP) was monitored with a Tono-pen XL tonometer several times prior to injection, and twice per week after injection. The basic IOP in each group served as a control. RESULTS: Experimental glaucoma occurred in 11 of 12 eyes in group 1 and IOP lasted from 20-50 days. The mean IOP levels were 29-35 mmHg (1 mmHg = 0.133 kPa), and the peak IOP levels were 37-45 mmHg. The IOP in two of ten eyes in group 2 increased to 22-50 mmHg and lasted for three days. The IOP of one of ten eyes in group 3 increased to 25-40 mmHg and lasted for four days. The IOP in the group 4 eyes increased by only a mean of 3 mmHg and lasted for one week. According to the standard, IOP above 22 mmHg for one week for successful experimental glaucoma, groups 2 and 3 were not ideal, and group 4 was a failure. CONCLUSIONS: An experimental glaucoma model induced by carbomer has the advantage of producing moderated and sustained IOP elevation. The model is simple to perform. It is simple, reliable, and useful for investigating optic nerve and retinal damage in glaucoma and for testing the toxicity and efficacy of various therapies.LA: Chinese
