Experimental researches of ocular hypertension retinal injury and the NOS mechanism in the injury

OBJECTIVES: (1) To observe the expression of NOS in the retina; (2) to investigate the function of the NOS inhibitor L-NAME (N<sup>G</sup>-nitro-L-arginine ester) in ischemia reperfusion of ocular hypertension. METHODS: The authors examined the expression of nNOS, eNOS, and iNOS in retinal ocular hypertension ischemia and reperfusion, and the affect of NOS inhibitor L-NAME on the expression using the immunohistochemistry SABC method. RESULTS: 1. nNOS, eNOS, and iNOS positive expression could be examined in retinal ganglion and neuroglial cells in ischemia reperfusion of ocular hypertension. nNOS was weakly positive in the normal control group. 2. After L-NAME injection, only iNOS was positive in the retinal ganglion and neuroglial cells, while nNOS and eNOS were negative in ischemia reperfusion of ocular hypertension. CONCLUSIONS: 1. NOS plays some cytotoxicity roles in the retina. 2. By inhibiting the activity of nNOS and eNOS, L-NAME may protect the retina against ischemia reperfusion injury.
