Effect of TGF-β1 on MMPs expression in cultured bovine trabecular cells

OBJECTIVE: To understand the effect of transforming growth factor-β1 (TGF-β1) on the expression of matrix metalloproteinases (MMPs) in cultured bovine trabecular cells (BTCs) and approach to pathogenesis and mechanism of primary open angle glaucoma (POAG). METHODS: BTCs were primarily cultured and subcultured. The third passage cells were incubated with different dosages of TCF-β1 (0, 0.5, 1, and 5 ng/ml, final concentration, diluted by DMEM) for 48 hours, MMP3 and MMP9 were determined by the immunohistochemistry method. The results were analyzed by a computer image-analysis system and statistical test. RESULTS: The cultured BTCs expressed MMP3 and MMP9. In comparison to the control group, TCF-[31 TGF-β1 could decrease the expression of MMP3 and MMP9 in BTCs. CONCLUSIONS: To a certain extent, TCF-β1 can decrease the expression of MMP3 and MMP9 in cultured BTCs, aggravate the stacking in ECM, and increase the resistance of aqueous outflow in the trabecular meshwork.LA: Chinese
