AIM: To study the effect of anandamide (AEA) on the expression of PGE2 and cytoskeleton in cultured bovine trabecular meshwork cells (BTM cells). METHODS: BTM cells were primarily cultured and subcultured. The third generation BTM cells were incubated with different dosages of AEA (0, 0.1, 1.0, 10μ mol/L, final concentration, diluted by DMEM) for 24 hours. Then the supernatant was collected. The morphology of BTM cells was observed by inverted microscope and the area of cells was calculated by computer image analytic system. The concentration of PGE2 in the medium was measured by the ELISA method. RESULTS: Compared with the control group, AEA could obviously induce the relaxation of BTM cells in a concentration-dependent manner. Moreover, in a certain range AEA can increase the expression of PGE2 in a dose-dependent manner. CONCLUSION: AEA may cause the relaxation of TM cells and promote the release of PGE2. LA: Chinese
