- Using a model of open and pressure-controlled culture system, it
was found that the increased pressure caused increasing in TGF-b2
excretion of cultured human trabecular meshwork (TM) cells. The
abnormal excretion of TGF-b2 may be one of the important mechanisms
for the pathological progression of POAG.
- In vitro, recombinant adeno-associated virus (rAAV)
vector mediated brain-derived neurotrophic factor (BDNF) transfected
cultured rat retinal ganglion cells effectively. The transfected
cell could express exogenous BDNF gene, which promoted their
activity and alleviated their apoptosis.
- Specific small interference RNA (siRNA) can interfere the
transcription and translation of Inhibitor of Kappa B kinase
b (IKK-b) and
inhibit the proliferation of the cultured fibroblasts of Tenon's
capsule in vitro.
- In an experimental cynomolgus monkey model of advanced glaucoma,
in the lateral geniculate nucleus (LGN) receiving input from the
glaucomatous eye, significant neuron loss and atrophy and
significant astroglial proliferation was observed; a certain degree
of neuron atrophy and gliocyte reactivity were also present in each
LGN layer connected to the unaffected fellow eye.
- For the same degree of structural damage (reduction in RNFLT),
the increase in VF scores is more dramatic in the advanced stage of
the disease compared to that in the early stage. Therefore, the
current Advanced Glaucoma Intervention Study (AGIS) and the
Collaborative Initial Glaucoma Treatment Study (CIGTS) scoring
systems are considered to be less sensitive to detect progression in
early glaucoma, since the steps for progression are defined
independently of the stage of the disease.
- In the early PACG, most of the Retinal Nerve Fiber Layer
Deficiency (RNFLD) was localised, while, in the early POAG, more
diffuse RNFLD could be found. In a pilot study of subjects with
early POAG, the inferior hemifield was more severely affected than
the superior. This was less pronounced in subjects with early PACG.
- Optic cup retinal stem cells (OC-RSCs) were isolated and
cultured successfully from the cup of embryonic rat E12.5. RA and
VEGF can be used as effective inducers to promote OC-RSCs'
differentiation.
- Laser peripheral iridotomy (LPI) can open 80% occludable angles
effectively in natural population.
- Dexamethasone can upregulate the expression of claudin-1 in
human trabecular meshwork cells, therefore, it can change the tight
junctions between cells and increase the filtration resistance. It
may be one of the reasons of elevated IOP in Steroid-Induced
Glaucoma.
- MA could significantly degrade the actin microfilaments in human
trabecular meshwork cells and could decrease IOP of rats and
monkeys. Its degradation action on the actin microfilaments in human
trabecular meshwork cells may become a potential anti-glaucoma
therapy.
