Expression and localization of ciliary neurotrophic factor mRNA in retina and optic nerve in rats

AIM: To investigate the effect of ciliary neurotrophic factor(CNTF) in protecting neuron induced by retina and optic nerve injury, and the correlation of the expression and localization of CNTF mRNA in normal retina and optic nerve in adult rats. METHODS: Frozen sections of eyeball and optic nerve in eight SD rats were made. In situ hybridization was conducted on the tissue sections by oligonucleotide probe marked by cardiox. The expressions and localization of CNTF mRNA were detected in retina and optic nerve. RESULTS: The result of in situ hybridization showed that the positive expression of CNTF mRNA in the normal retina tissue was in inner nuclear layer. The positive cells of CNTF mRNA in the normal optic nerve were in a beaded arrangement along the long axis. CONCLUSION: The positive expression of CNTF mRNA in the normal retina tissue of adult rats is in inner nuclear layer, and the CNTF mRNA has a comprehensive expression in the optic nerve. The characteristics of distribution may have the relation with the fact that CNTF can promote the survival and axonal regeneration of injured retina ganglion cells. LA: Chinese
